Cytotoxicity of Epitope-specific CD8+ T Cells

Activation of a robust CD8+ T cell response should be considered when developing and designing vaccines for intracellular infections and cancer therapies. CD8+ T cells can directly kill infected and transformed cells in cancer and are directly associated with beneficial protection in many disease models. In the field of drug analysis, cytotoxicity and proliferation assays are commonly used in drug screening to detect whether test molecules have an effect on cell proliferation or show direct cytotoxic effects. The purpose of the detection method under the purpose of drug testing is to confirm the number of remaining viable cells. It includes a variety of essays based on various cellular functions, such as enzymatic activity, cell membrane permeability, cell adhesion, ATP production, coenzyme production, and nucleotide uptake activity. While T cell-mediated cytotoxicity requires assessment of the self-activity and lethality of cytotoxic T cells. Cytotoxic T cells are an important part of the adaptive immune system and are essential for clearing infections and fighting tumors. Tumors often have multiple mechanisms for evading T cells, which has prompted researchers to enhance the understanding of the role of T cells in anti-tumor immunity, and tumor-killing assays can assess the ability of T cells in immunotherapy.

Cytotoxicity Assay of Epitope-specific CD8+ T Cells

Our service is designed to help customers test the function of molecules of interest, and the core of this service is T cell activity analysis. We offer our clients a variety of methods to measure CD8+ T cell cytotoxicity, including in vitro and in vivo assays.

  • In Vivo Assay
    To determine the ability of CD8+ T cells to kill their targets at their cognate receptors and their natural environment, in vivo cytotoxic T cell assays (in vivo antigen-specific CTLs) are ideal. In vivo, CTLs assays provide a snapshot of the overall ability of the host to kill "target" cells by measuring the loss of incoming target cells relative to "non-target" cells. In vivo services enable better assessment of CD8+ T cell cytotoxicity to their cognate targets. Fluorescence-based labeling methods have higher sensitivity and better safety and allow for specific labeling. We can then complete tracking and analysis through imaging techniques.
  • In Vitro Assay
    In vitro assays have broader applications and we have a range of cell killing assay options. Our CD8+ T cytotoxicity assays include methods to measure IFNγ production by ELISPOT or intracellular cytokines or cytotoxic particles by flow cytometry. In addition to cytotoxic T cell assays, we also offer Natural Killer (NK) cell assays. In addition, we also offer assays based on flow cytometry and live-cell imaging. The choice of these analysis methods can be chosen according to your project.
