Most T lymphocytes express clones and highly specific antigen receptors (TCRs) on the cell surface. TCR (T cell receptor) binds to MHC peptide complex and recognizes antigenic peptides presented in MHC molecular environment. Because TCR has low affinity for MHC peptide complex and fast dissociation rate, the recombinant soluble monomer MHC peptide complex has not been used to detect antigen-specific T cells.
Tetramer analysis is used to detect and quantify antigen-specific T cells. The interaction between peptide MHC complex (pMHC) and homologous T cell receptor (TCR) is too low to be detected in flow cytometry. The development of tetramer MHC peptide complexes has led to the direct visualization, quantification and phenotypic characterization of antigen-specific T cells, which greatly enhances our ability to evaluate cell dynamics during immune response. The multicomponent polymerization of monomeric biotinylated pMHC by streptomycin complexation increases the desire of pMHC streptomycin tetramer for homologous TCR in order to detect antigen-specific T cells.
MHC tetramer technology is based on the ability of MHC peptide complexes to recognize antigen-specific T cells at the single-cell level. This breakthrough technology enables researchers to accurately measure targeted T cell responses in infectious diseases, cancer and autoimmune diseases.
