T Cell Activation and Cytokine

T cells generated in the T thymus are programmed to act as bodyguards for specific antigens. After leaving the thymus, T cells circulate throughout the body until their antigens are recognized on the surface of antigen-presenting cells (APCs). TCRs on CD4⁺ helper T cells and CD8⁺ cytotoxic T cells bind to the antigen, triggering initial activation of T cells. This initial binding between T cells specific for one antigen and its matching antigen-MHC initiates the overall response. CD4⁺ T helper cells play a key role in adaptive immunity. They regulate the functions of other important immune cells, such as B cells, macrophages, and CD8⁺ cytotoxic T cells, through cytokine secretion. The key to activating CD4⁺ T cells is the specific recognition of pMHC ligands displayed on the surface of antigen-presenting cells (APCs) through their αβTCRs. Antigen recognition triggers various intracellular signaling events that affect the entire immune response, so cytokine analysis becomes important.
