T cells express T cell receptors (TCR) that recognize short peptide antigens in the context of major histocompatibility class (MHC) molecules. This recognition process generates a series of cytokine-mediated signals that help control the immune response. Modification of MHC-associated peptides is desirable for the design of vaccines containing unnatural, biostable MHC peptides. Effects of peptide backbone modification on T cell mediates recognition of tumor-associated antigens bound to MHC.

Peptides are modified in various ways, such as synthesized as biotin, fluorescent dyes, phosphorylation, methylation, acylation, acetylation, unnatural amino acids, etc. Modifications can also be classified as N-terminal modifications, C-terminal modifications, side chain modifications, amino acid modifications and backbone modifications. MHC-associated peptide modification is mainly based on changing the main chain structure or side chain motifs as an important way. The effect of such modifications on MHC affinity and T-cell activation was measured. Many of these modifications can reduce or enhance T-cell responses.
