Introduction of Peptide Arrays for Epitope Mapping

Peptide arrays are collections of peptides displayed on a solid surface, usually have a slide or membrane. Peptide arrays with immobilized peptides are used to study the binding properties, function and kinetics of protein-protein interactions, a key technique for mapping antibody epitopes and finding protein-bound residues in studies related to MHC. Since the development of therapeutics relies on detailed knowledge of these structural domains, peptide libraries are used to display multiple short, linear peptide fragments and derive specific epitopes in parallel. The most common applications for epitope mapping are T cells and antibodies.

Peptide arrays show overlapping peptide scans by MHC delivery of antigen or any other peptide set. They can be incubated with antibodies, serum or other patient samples. For example, peptides from viral or bacterial antigens, overlapping peptides from tumor-associated antigens, collection of antigens from specific pathogens or cancer types, can all be useful for MHC-associated immunotherapy studies.
