Introduction of MHC Proteins

The detection of antigen-specific T-cell responses by MHC multimer staining is rapidly becoming one of the core immunology techniques, and the production technology of MHC multimers has been greatly optimized in recent years. In addition, recent work has demonstrated the potential for high-throughput detection of T-cell responses and demonstrated that manipulation of T-cell responses is also feasible through the use of multimeric MHC reagents. Allelic polymorphisms in T-cell receptors have profound effects on immune responses, especially epitope-specific TCRs that are present in specific diseases often exhibit strong population bias. Studies have found the impact of overlooked sequence variants in TCRs on immunity by MHC pentamer staining analysis. This is due to the fact that a single amino acid difference within the TCR may reduce its binding affinity for the viral peptide-MHC complex and render this population of cells less dominant in immune responses.
