MHC-associated Peptide Pool for Antigen Target Discovery

Creative BioMart offers the combination of MHC alleles and peptide pools, highlighting T cell reactivity in antigen target discovery of samples. Because the frequency of any particular MHC allele can be elucidated, this type of analysis is expected to be a valuable method for estimating the likelihood of T cell responses within a group.

Introduction of Peptide Pool for Antigen Target Discovery

The peptide library contains a variety of restricted epitopes of MHC class I or class II molecules from human viruses. Most patients who have been exposed to these antigens and possess the corresponding CD8+ and CD4+ memory T cells. This means that, unlike conventional reagents, the MHC-associated peptide pool is well suited for use as a specific positive control for all T cell-based assays, such as intracellular flow cytometry analysis or ELISPOT assays.

T cell activation assays rely on donors that are exposed to specific proteins or peptides for uptake by antigen presenting cells (APCs) and eventual presentation of MHC-treated peptides to T cells. Several methods can be used to analyze the T cell adaptive immune response against the protein. Some of these assays include ELISPOT for the detection of secreted cytokines and immunostaining for intracellular cytokines and markers of cell membrane activation.

Our Peptide Pool for Antigen Target Discovery

We used synthetic overlapping peptide libraries with multiple sequences for T cell activation assays. Each library consists of several short peptides with overlapping amino acids. After incubation of the donor source with each peptide library, T cell activation and target discovery were determined based on positive immunostaining.
