MHC II/Peptide Affinity Assays

Creative BioMart offers MHC/peptide affinity analysis services for MHC class II alleles. This service is used to determine the most likely immunogenic peptides in protein sequences, aiding the development of novel immunotherapies and therapeutic biological proteins. Our technical services are widely applicable to infectious disease research, vaccine development and other fields. Our platform provides methods for immunogenicity screening using MHC II. Current research shows that more than 80% of cancer-related neo-epitopes may be involved in antigen presentation by MHC II, so MHC II is increasingly included in cancer vaccine development.

MHC Class II Molecular Binding Assay

The purpose of binding assays is to measure the interaction between two molecules, such as a protein and another protein. This has an important place in the development of biotherapeutic protein formulations. Design flaws in binding experiments can limit the information obtained, especially if the affinity of the reactants for each other is not measured. The binding determination of MHC II molecules and peptides broadly includes the determination of equilibrium constants and the determination of stability. The number of equilibrium constants characterizes the affinity between molecules, while stability is another aspect. Although a wide range of binding prediction algorithms exists for peptide binding to MHC class II alleles, the accuracy of these methods does not always correlate well with physical binding data. MHC class II affinity analysis of peptides overcomes these limitations by determining whether peptide sequences bind to MHC class II molecules, avoiding the false positive and false negative results of algorithmic approaches.

Service Process

Our MHC Peptide Complex Affinity Analysis Services for Analytical Testing Platforms provide the following services for binding analysis of MHC class II molecules and peptides in exogenous antigen presentation and cross-presentation. This service is available for MHC class II genes in customer samples and is available for HLA-DR, HLA-DR and HLA-DR alleles, and some mouse class II alleles. Our services include the design of custom MHC monomers or tetramers or the selection of commercialized proteins, and the design of custom synthetic peptides is done by our scientists. Candidate molecules screened by the HLA peptide binding assay can be further analyzed for assay rate or stability. Analysis of on and off rates in binding assays will provide kinetic information on the affinity properties of individual peptides. Our suppliers can provide products for identification and HLA-DR, DP or DQ, an important part of ensuring the quality of service.
