Major histocompatibility complex (MHC) molecules are known for their role in antigen (cross) presentation and thus play a key role in communication between immune cells. However, MHC molecules can also act as signal receptors and are involved in what is known as reverse MHC signaling, and the linkage of MHC molecules can lead to signal transduction and cellular regulation in antigen-presenting cells. In the case of MHC-I, for example, reverse signaling can produce a variety of outcomes, including apoptosis, migration, induction or reduction of proliferation, and cytotoxicity to target cells. MHC-I stimulates signaling pathways and signaling molecules such as Akt1, extracellular signal-regulated kinase 1/2 (ERK1/2) and nuclear factor-κB (NF-κB) in various immune and non-immune cells. Common signaling molecules activated upon MHC-I ligation in a variety of cell types encompass these molecules. Natural MHC-I ligands and extended downstream pathways are not fully understood, but reverse MHC-I signaling appears to be involved in regulating defenses against bacterial and viral infections and malignancies. The study of MHC signaling is therefore important for understanding its impact on disease physiology.
