Folding and transport of proteins are tightly regulated in all cells through the endoplasmic reticulum, such as major histocompatibility complex (MHC) class I. MHC overexpression plays an important role in the pathophysiology of some diseases. Taking protein-rich muscle tissue as an example, overexpression of MHC class I proteins, a common feature of many myopathies, including idiopathic myositis, induces ER stress. Studies have investigated the consequences of MHC overexpression in muscle at different ages. Using a transgenic model of myositis induced by MHC class I protein overexpression, it found that the younger group had a more severe disease phenotype, with rapid onset of muscle weakness and pathology. Of course, researchers can also use in vitro overexpression constructs, and the obtained custom proteins can be used to study the pathological or physiological changes caused by MHC overexpression.
