To monitor antigen-specific T cell responses, antigen-specific T cells can be stimulated with MHC-related peptides or antigens to analyze proliferation and cytokine production. Flow cytometry readout can be used for intracellular cytokine staining and is the basis for a variety of assays developed to characterize immune responses by multicolor flow cytometry. It can be combined with cell surface markers for immunophenotypic analysis to measure T cell population sensing.
Like MHC multimer staining, this is a method that allows direct measurement of antigen-specific T cells. However, it has requirements that depend on measuring the same activated cells as the antigen-specific cells. One of its advantages is the co-staining of T cell lineage markers and possibly other surface or intracellular markers (e.g. involved in maturation, suppression or activation), thus increasing the level of knowledge obtained. The combination of flow cytometry and MHC protein staining allows measurement of cytokine expression and provides additional information about the immune response.
