MHC Class I Molecular Binding Assay

MHC class I molecules are often associated with the presentation of endogenous antigens, and rapid epitope discovery enables the screening of T-cell epitopes of any protein in weeks rather than months or years. MHC class I molecules are present in all vertebrates and control cellular immune responses against viruses and other intracellular pathogens. Complexes are generated when antigenic peptides and MHC class I molecules efficiently interact, which are displayed on the surface of presenting cells and are targeted by peptide-specific, MHCrestricted cytotoxic T cells. If the peptide is of foreign origin, T cells may attack and lyse the target cell. It is important to be able to predict and measure the affinity of peptide-MHC binding. MHC I affinity analysis can significantly speed up the T-cell epitope discovery process and overcome challenges associated with time-consuming, labor-intensive functional assays and limited patient sample sizes. Service Process

Our MHC peptide complex resolution service is used for the analysis of the binding affinity of peptides bound to MHC-I. Although accurate and reliable computational methods have been developed to predict the binding affinity of peptides for binding to MHC-I, these methods have had great success in narrowing the search for T cell epitopes. But its high false positives for immunogenicity (i.e., only a few of the peptides predicted and subsequently shown to bind to MHC-I are actually recognized by T cells after infection) make binding-test-based affinity assays of great potential. The pipeline for this service includes custom peptide synthesis, quantitative binding of candidate peptides to MHC alleles, and binding rate analysis. The MHC peptide binding assay can rapidly determine the ability of each candidate peptide to bind to one or more MHC alleles compared to control peptides.
