MHC Tetramers Production

Creative BioMart offers customization services for a variety of MHC/peptide complex products, including customization of MHC tetramers. The pMHC tetramer is composed of 4 MHC class I or class II complex monomers loaded with specific antigenic peptides bound to fluorescently labeled streptavidin molecules. Our tetramers can be used in a wide variety of tests of T cell function.

Introduction of MHC Tetramers

The major histocompatibility complex (MHC) is a family of proteins present in most vertebrates that encode a group of membrane glycoproteins called MHC molecules. The main function of MHC molecules is to participate in the collective immune response, present antigenic peptides to target cells, and affect the homeostasis of the immune system. An MHC tetramer is an MHC/peptide complex, which is a single (monomeric) MHC molecule bound to its cognate peptide at a site remote from the T cell recognition site. This monomeric complex is capable of binding biotin at 4 different sites to form a tetrameric complex known as the MHC tetramer. Methods to define the type of antigen-specific T lymphocyte response by characterizing and characterizing the immune response have certain limitations. In order to accurately identify the type of T lymphocyte response, the MHC tetramer (tetramer) technology, which can directly label antigen-specific T lymphocytes, has been gradually developed. Tetramer technology increases the sensitivity of specific T cell detection.

Our MHC/Peptide Tetramer Production

We provide the preparation services of MHC peptide complex and its tetramer, from MHC protein and the polypeptide folded to form a monomer complex, and then the complex monomer is labeled with biotin tetramerized by the interaction of biotin and Streptavidin.

Application Fields

MHC tetramer has been proved to accurately and effectively monitor T cell response, and it is widely used in the following fields.

  • Pathogens and Infections

T cells play a key role in the adaptive immune response to pathogen infection. Our custom-made MHC tetramers have been shown to be very efficient in identifying antigen-specific T cells by flow cytometry, even those that occur at low frequencies in freshly sampled lymphocyte populations..

  • Cancer

Our MHC tetramers designed for you allow ex vivo selection and proliferation of tumor-associated antigen-specific T cells, which are heavily influenced in cancer immunotherapy, and then be reintroduced to boost the immune system.

  • Others

Applications of our proven MHC tetramer technology are expanding to include analysis of T cells in adaptive immune responses against parasites, transplanted tissues, allergens, and even self-antigens. Our products add new possibilities for uncovering potential therapeutic targets and developing biotherapeutic peptides.
